Processes leading to acute myeloid leukemia discovered

ScienceDaily (Jan. 30, 2012) ? Researchers at UC Santa Barbara have discovered a molecular pathway that may explain how a particularly deadly form of cancer develops. The discovery may lead to new cancer therapies that reprogram cells instead of killing them. The findings are published in a recent paper in the Journal of Biological Chemistry.

The UCSB research team described how a certain mutation in DNA disrupts cellular function in patients with acute myeloid leukemia (AML). The researchers were prompted to study this process by another research team's discovery that AML patients have a mutation in a certain enzyme, which was reported in the New England Journal of Medicine. The enzyme is a protein called DNMT3A, which leads to changes in how the DNA of AML patients is methylated, or "tagged." Norbert Reich, professor in the Department of Chemistry and Biochemistry at UCSB, was already studying that particular enzyme with his research group, so they began to study the disease process of AML at the cellular level.

Reich explained that tagging is a way of reading DNA at the cellular level. This falls within an area of study called epigenetics, a process that occurs "on top" of genetics. Each person has approximately 200 types of cells, all with the same DNA, and these must be controlled in different ways. "There is an enzyme -- a protein -- that tags DNA and controls which of the genes in your cells, your DNA, gets turned on and off," said Reich. "So you have 20,000 genes, and you have to control them differently in your brain than in your liver."

Reich explained that there is current interest in this broader field of epigenetics as a direction for the treatment of cancer. "There's definitely the idea that this may be a new way of developing therapeutics, because you don't have to kill the cancer cell," said Reich. "Almost every cancer therapy that's out there works on the principle that a cancer cell needs to be killed."

With epigenetics, instead of only having DNA sequence coding for certain genes, there is an epigenetic process, with another layer of information on top of the genetic process. In this case, that information is the tagging by the methyl groups.

"If you really think about it, this is part of the answer as to how your cells can be so different and yet they all have the same DNA," said Reich. "You have the same genome in every one of your cells, but you do not have the same epigenome, which is basically the methylation pattern, the tagging pattern. That is different in every type of your cells. And the way this relates back to cancer, with leukemia, in those patients, the tagging is messed up. The patterns are not correct. Our big contribution to that is we've explained how the mutations in the enzyme could lead to that disruption of the tagging pattern."

The UCSB group developed a test to demonstrate that the mutant enzymes in AML can only work on DNA for short distances. As a result, the precise methylation patterns of a healthy cell are disturbed, resulting in genes being turned on at the wrong place and time, which in turn can initiate the growth of cancerous cells.

The team found that the mutation AML patients have causes a certain complex of four proteins to be disrupted. "The surprise was that the disruption doesn't stop the enzyme from being active; it doesn't stop the enzyme from tagging the DNA," said Reich. "Instead, it stops the way it can do it. Instead of going to your DNA and tagging an entire region of chromosome, it goes there, does one thing, and leaves. That process, that change, is what we see in the AML patients. So we think we have a molecular explanation for this disease."

Reich said that the currently prescribed drug Vidaza works by affecting the same enzyme that is mutated in AML. There is interest in the pharmaceutical industry in developing other therapeutics to target the enzymes responsible for tagging the DNA. These epigenetic inhibitors would reprogram rather than kill the cell.

Traditional cancer therapies use radiation and chemotherapy to remove or kill cancer cells. "The problem with that is that cancer cells are often very subtly different from normal cells," said Reich. "So you have one of the most difficult therapeutic challenges known to man, which is to distinguish between two human cells -- one that's cancerous and one that's not. Instead of killing the cell, the notion is that if you could just reprogram the cell, then it goes back to being normal. You intercept the cancer development. This is still an aspiration; it hasn't been achieved really, but that's what attracts people to the field of epigenetic-based therapies, because of the prospect of not having to kill cells."

Celeste Holz-Schietinger and Douglas Matje, both graduate students working in the Reich lab, are the first and second authors of the paper.

Is Alec Baldwin hinting at the end of '30 Rock'?

After Alec Baldwin gleefully picked up his sixth straight Screen Actors Guild Award for his role as "30 Rock" boss Jack Donaghy on Sunday,?he was already busy pitching himself to another sitcom.

According to the LA Times blog, Baldwin told fellow nominees Ty Burrell and Eric Stonstreet of "Modern Family," "You know my show is almost over ... I'm actually pitching myself to you guys."

But is his show almost over?

Last fall NBC negotiated a two-year contract for him that would keep him on the show this season and next?-- if the sitcom continues.

(FYI, TODAY.com is powered by msnbc.com, which is a joint venture between Microsoft and NBC Universal.)

The 2011 season finished with "30 Rock" almost even in the ratings with the now-canceled "Outsourced," and this season has not seen a dramatic jump in viewers for the sitcom. Indeed, NBC head of entertainment, Robert Greenblatt, inherited a shaky house when he took over the reins about a year ago, and he knows he needs to shape up the schedule to try to?improve the network's ratings.

But could nuking "30 Rock" early be part of that restructuring?

Greenblatt told TODAY.com he hasn't decided yet if he even wants the comedy on his schedule. "I'll decide by the May upfront," he told us.

It's possible the show -- which has so far survived thanks to its numerous awards and critical praise -- may not have the needed viewers to stay on the schedule for the full renewal.

One thing's for sure: Greenblatt seems to have a?limited tolerance for low performers. When asked why he opted to chop "Chuck" and burn off the episodes quickly, including last Friday's double-header finale, he told us, "Have you seen the ratings on 'Chuck? Those rabid fans going crazy on the net didn't come to church. 'Chuck's' time had come."

"Chuck's" finale averaged 4.2 million viewers on Friday, while "30 Rock" grabbed 4.1 million in its timeslot Thursday.

Time short for Gingrich to close gap in Florida (AP)

MIAMI ? Newt Gingrich slammed GOP rival Mitt Romney on Sunday for the steady stream of attacks he likened to "carpet-bombing," trying to cut into the resurgent front-runner's lead in Florida in the dwindling hours before Tuesday's pivotal presidential primary.

Surging ahead in polls, Romney kept the pressure on Gingrich with a dominant advertising presence that questioned the former House speaker's leadership and ethics. During campaign stops, Romney divided his focus between Gingrich and President Barack Obama.

In what has become a wildly unpredictable race, the momentum has swung back to Romney, staggered last weekend by Gingrich's victory in South Carolina. Romney has begun advertising in Nevada ahead of that state's caucuses next Saturday, illustrating the challenges ahead for Gingrich, who has pledged to push ahead no matter what happens in Florida.

Romney's campaign has dogged Gingrich at his own campaign stops, sending surrogates to remind reporters of Gingrich's House ethics probe in the 1990s and other episodes in his career.

Gingrich reacted defensively, accusing the former Massachusetts governor and a political committee that supports him of lying, and the GOP's establishment of allowing it.

"I don't know how you debate a person with civility if they're prepared to say things that are just plain factually false," Gingrich said during appearances on Sunday talk shows. "I think the Republican establishment believes it's OK to say and do virtually anything to stop a genuine insurgency from winning because they are very afraid of losing control of the old order."

Gingrich objected specifically to a Romney campaign ad that includes a 1997 NBC News report on the House's decision to discipline Gingrich, then speaker, for ethics charges.

After hounding Gingrich during two debates last week, Romney returned more of his attention to Obama, who had been Romney's chief target as he tried to make the case that he was the most worthy Republican to challenge the Democratic incumbent.

But Romney didn't relent in swiping at Gingrich, even as an NBC News/Marist poll published Sunday showed Romney with support from 42 percent of likely Florida primary voters, compared with 27 percent for Gingrich.

"He's now finding excuses ... complaining about what he thinks were the reasons he thinks he's had difficulty here in Florida. But you know, we've got a president who has a lot of excuses," Romney said at a rally in Naples. "And the excuses are over, it's time to produce."

Former Pennsylvania Sen. Rick Santorum, trailing in Florida by a wide margin, stayed in his home state, where his 3-year-old daughter, Bella, was hospitalized. She has a genetic condition caused by the presence of all or part of an extra 18th chromosome. Aides said he would resume campaigning as soon as possible.

Texas Rep. Ron Paul, who has invested little in Florida, looked ahead to Nevada. The libertarian-leaning Paul is focusing more on gathering delegates in caucus states, where it's less expensive to campaign. But securing the nomination only through caucus states is a hard task.

The race began moving toward a two-person fight in South Carolina, and has grown more bitter and personal in Florida.

The intense effort by Romney to slow Gingrich is comparable his strategy against Gingrich in the closing month before Iowa's leadoff caucuses Jan. 3.

Gingrich led in Iowa polls, lifted by what were hailed as strong performances in televised debates, only to drop in the face of withering attacks by Romney, aided immensely by ads sponsored by a political committee run by former Romney aides.

In Florida, senior Romney aides have popped up at Gingrich events to question Gingrich's conservative credentials. Led by Romney's top Iowa adviser, David Kochel, Romney's team cites Gingrich's criticism of House Budget Committee chairman Paul Ryan's Medicare overhaul plan last year, and his appearance with then-House Speaker Nancy Pelosi, D-Calif., in an advertisement supporting climate-change legislation.

"That kind of language emboldens the critics of conservatism," Kochel said. "We're out pointing that out correcting the record."

Gingrich has responded by criticizing Romney's conservative credentials. Outside an evangelical Christian church in Lutz, Gingrich said he was the more loyal conservative on key social issues.

"This party is not going to nominate somebody who is a pro-abortion, pro-gun-control, pro-tax increase liberal," Gingrich said. "It isn't going to happen."

But Gingrich, in appearances on Sunday news programs, returned to complaining about Romney's tactics, rather than emphasizing his own message as that of a conservative with a record of action in Congress.

"When we get to a positive idea campaign, I consistently win," Gingrich said. "It's only when he can mass money to focus on carpet-bombing with negative ads that he gains any traction at all."

Romney and the political committee that supports him had combined to spend some $6.8 million in ads criticizing Gingrich in the Florida campaign's final week. Gingrich and a group that supports him were spending about one-third that amount.

Gingrich worked to portray himself as the insurgent outsider, collecting the endorsement of tea party favorite Herman Cain, whose own campaign for president foundered amid sexual harassment allegations.

It was unclear how aggressively Gingrich would be able to compete in states beyond Florida. The next televised debate, a format Gingrich has used to his advantage, is not until Feb. 22, more than three weeks away.

Romney already has campaigned in Nevada more than Gingrich, is advertising there, and stresses his business background in a state hard-hit by the economy. His campaign welcomed the Sunday endorsement of the Las Vegas Review-Journal, Nevada's largest newspaper.

Michigan and Maine, states where Romney is well-positioned also hold their contests in February. Arizona, a strong tea-party state where Gingrich could do well, has its primary Feb. 28.

Fire kills 26 patients at rehab center in Peru (Reuters)

LIMA (Reuters) ? Fire swept through a rehabilitation clinic for drug addicts and alcoholics in Peru on Saturday, killing 26 patients who were locked in to stop them from fleeing during treatment, local media and witnesses said.

Several survivors said the blaze was started by two patients who wanted to break out of the private "Christ is Love" center in the capital Lima.

"Some inmates wanted to escape and they set fire to some cloths and started throwing them at the manager's office, and then it caught fire inside," said patient Wilmer Garcia.

Most of the victims were thought to have died from smoke inhalation.

Lima Mayor Susana Villaran said the center lacked a municipal licence and that "drastic action" was needed to prevent similar incidents in the Andean country.

"This is a great tragedy," she told local radio. "Families hand over relatives who are suffering drug and alcohol addiction so they can be rehabilitated and often they're desperate. These places must be supervised by the Health Ministry and municipal authorities."

Firefighters and prosecutors have opened investigations into the cause of the fire.

Obama's Congressional Reform Proposals Will Never Pass (ContributorNetwork)

COMMENTARY | President Barack Obama is proposing sweeping changes in the way in which Congress works, according to the Associated Press. His reforms would include policies against insider trading in Congress, bundled campaign contributions, restrictions on stocks and securities government officials could own and changes to appointment approvals. As a political scientist, I see this as a political plot against Congress.

The president is making suggestions that people would automatically agree with. Personally, I think many of his suggestions would clear up corruption in Congress and would clear up many of the campaign donations that can cause corruption. The fact is, his proposals would never pass as is and he knows it. He is making a suggestion to change how politics work behind the scenes, and Congress will not allow the "Big Boys Club" to lose their toys.

Few in Congress would support President Obama's proposed changes for the right reason. I could see the "supporters" of the sweeping changes as doing it for show as they know the changes would never pass. The Republicans are automatically going to oppose the bill for the public reason of it "being too restrictive and an attack on Congress." The bill would die and the "corruption of the Republican Party" would be blamed.

Congress has very low approval ratings and President Obama has been bumping heads with the body at every turn. Why not try to get some political points off of the group nobody likes or trusts these days? At the same time, the president can appear to be someone who is looking out for the best interests of the common person and of the government as a whole.

Over the next few weeks, we are sure to see this fight play out in the media. President Obama will press how Congress is wrong for not passing the changes immediately. The Democrats will accuse the Republicans of being corrupt, while the "Blues" try to hide the corruption which they are guilty of. Of course, the Republicans will be working on covering up their corruption while pushing the idea of the president "grandstanding" and being a "populist." All the president has to hope is that Congress does not push back by revealing the skeletons in his closet.

29 Chinese missing after militant attack in Sudan (AP)

BEIJING ? Militants apparently captured 29 Chinese workers after attacking a remote worksite in a volatile region of Sudan, and Sudanese forces were increasing security for Chinese projects and personnel there, China said Sunday.

China has close political and economic relations with Sudan, especially in the energy sector.

The Foreign Ministry in Beijing said the militants attacked Saturday and Sudanese forces launched a rescue mission Sunday in coordination with the Chinese embassy in Khartoum.

The Ministry's head of consular affairs met with the Sudanese ambassador in Beijing and "urged him to actively conduct rescue missions under the prerequisite of ensuring the safety of the Chinese personnel," the statement said.

In Khartoum, a Chinese embassy spokesman said the northern branch of the Sudan People's Liberation Movement announced that 29 Chinese workers had been captured in the attack. The spokesman, who asked not be identified, gave no other details and it wasn't clear if the militants had demanded conditions for their return.

Other details weren't given. The official Xinhua News Agency cited the state governor as saying the Sudan People's Liberation Movement attacked a road-building site in South Kordofan and seized the workers.

The Sudan People's Liberation Movement are a guerrilla force that has fought against Sudan's regime. Its members hail from a minority ethnic group now in control of much of South Sudan, which became the world's newest country only six months ago in a breakaway from Sudan.

Sudan has accused South Sudan of arming pro-South Sudan groups in South Kordofan. The government of South Sudan has called such accusations a smoke screen intended to justify a future invasion of the South.

China has sent large numbers of workers to potentially unstable regions such as Sudan and last year was forced to send ships and planes to help with the emergency evacuation of 30,000 of its citizens from the fighting in Libya.

China has consistently used its clout in diplomatic forums such as the United Nations to defend Sudan and its longtime leader Omar al-Bashir. In recent years, it has also sought to build good relations with leaders from the south, where most of Sudan's oil is located.

Chinese companies have also invested heavily in Sudanese oil production, along with companies India and elsewhere.

In a study published in the journal Geology, scientists at the University of Miami (UM) Rosenstiel School of Marine and Atmospheric Science suggest that the large changes in the carbon isotopic composition of carbonates which occurred prior to the major climatic event more than 500 million years ago, known as 'Snowball Earth,' are unrelated to worldwide glacial events.

"Our study suggests that the geochemical record documented in rocks prior to the Marinoan glaciation or 'Snowball Earth' are unrelated to the glaciation itself," said UM Rosenstiel professor Peter Swart, a co-author of the study. "Instead the changes in the carbon isotopic ratio are related to alteration by freshwater as sea level fell."

In order to better understand the environmental conditions prior to 'Snowball Earth', the research team analyzed geochemical signatures preserved in carbonate rock cores from similar climactic events that happened more recently ? two million years ago ? during the Pliocene-Pleistocene period.

The team analyzed the ratio of the rare isotope of carbon (13C) to the more abundant carbon isotope (12C) from cores drilled in the Bahamas and the Enewetak Atoll in the Pacific Ocean. The geochemical patterns that were observed in these cores were nearly identical to the pattern seen prior to the Marinoan glaciation, which suggests that the alteration of rocks by water, a process known as diagenesis, is the source of the changes seen during that time period.

Prior to this study, scientists theorized that large changes in the cycling of carbon between the organic and inorganic reservoirs occurred in the atmosphere and oceans, setting the stage for the global glacial event known as 'Snowball Earth'.

"It is widely accepted that changes in the carbon isotopic ratio during the Pliocene-Pleistocene time are the result of alteration of rocks by freshwater," said Swart. "We believe this is also what occurred during the Neoproterozoic. Instead of being related to massive and complicated changes in the carbon cycle, the variations seen in the Neoproterozoic can be explained by simple process which we understand very well."

Scientists acknowledge that multiple sea level fluctuations occurred during the Pliocene-Pleistocene glaciations resulting from water being locked up in glaciers. Similar sea-level changes during the Neoproterozoic caused the variations in the global carbon isotopic signal preserved in the older rocks, not a change in the distribution of carbon as had been widely postulated.

One day in 2010, Rutgers physicist Vitaly Podzorov watched a store employee showcase a kitchen gadget that vacuum-seals food in plastic. The demo stuck with him. The simple concept an airtight seal around pieces of food just might apply to his research: developing flexible electronics using lightweight organic semiconductors for products such as video displays or solar cells.

"Organic transistors, which switch or amplify electronic signals, hold promise for making video displays that bend like book pages or roll and unroll like posters," said Podzorov. But traditional methods of fabricating a part of the transistor known as the gate insulator often end up damaging the transistor's delicate semiconductor crystals.

Drawing inspiration from the food-storage gadget, Podzorov and his colleagues tried an experiment. They suspended a thin polymer membrane above the organic crystal and created a vacuum underneath, causing the membrane to collapse gently and evenly onto the crystal's surface. The result: a smooth, defect-free interface between the organic semiconductor and the gate insulator.

The researchers reported their success in the journal Advanced Materials. In the article, Podzorov and three colleagues describe how a single-crystal organic field effect transistor (OFET) made with this thin polymer gate insulator boosted electrical performance. The researchers further reported that they could remove and reapply membranes to the same crystal several times without degrading its surface.

Organic transistors electrically resemble silicon transistors in computer chips, but they are made of flexible carbon-based molecules that can be printed on sheets of plastic. Silicon transistors are made in rigid, brittle wafers of silicon.

One day in 2010, Rutgers physicist Vitaly Podzorov watched a store employee showcase a kitchen gadget that vacuum-seals food in plastic. The demo stuck with him. The simple concept an airtight seal around pieces of food just might apply to his research: developing flexible electronics using lightweight organic semiconductors for products such as video displays or solar cells.

"Organic transistors, which switch or amplify electronic signals, hold promise for making video displays that bend like book pages or roll and unroll like posters," said Podzorov. But traditional methods of fabricating a part of the transistor known as the gate insulator often end up damaging the transistor's delicate semiconductor crystals.

Drawing inspiration from the food-storage gadget, Podzorov and his colleagues tried an experiment. They suspended a thin polymer membrane above the organic crystal and created a vacuum underneath, causing the membrane to collapse gently and evenly onto the crystal's surface. The result: a smooth, defect-free interface between the organic semiconductor and the gate insulator.

The researchers reported their success in the journal Advanced Materials. In the article, Podzorov and three colleagues describe how a single-crystal organic field effect transistor (OFET) made with this thin polymer gate insulator boosted electrical performance. The researchers further reported that they could remove and reapply membranes to the same crystal several times without degrading its surface.

Organic transistors electrically resemble silicon transistors in computer chips, but they are made of flexible carbon-based molecules that can be printed on sheets of plastic. Silicon transistors are made in rigid, brittle wafers of silicon.

In my first 'major' business - ie, major for me - I had a crew of 17 men for a hair under 20 years. We put work in a dozen countries. I detest policys, so we had very few of them - a total of 6 or 7. The ones we had made Sense. Such as: "There is no pecking order here. There's Carol & the rest of you. If you have a problem you can't work out on your own in adult manner, SEE THE BOSS so we can fix it." And: "You were hired because you are an adult. ADULT behavior is expected here." And - "WE are 'the company'. If you steal from 'the company', you steal from yourself as well as the rest of us. Do it enough & no one will have a job here." & - "If you come here with alcohol in you & I find out, you will be fired. If you bring alcohol here, I will kill you, resurrect you, & then fire you. If you come here with drugs in you, you will be fired. If you bring drugs on the premises, I will kill you, resurrect you, & repeat until I'm too tired to do it again, & then I will fire you. Never think I'm kidding about this." I told them all, "The building has electricity & heat 24 hours a day. If you like working 2 a.m. - whenever, that's up to you, so long as you get your work done to spec & on time. No visitors unless they're cleared ahead of time, and no shenanigans. Keep the door locked if you're working here during non-business hours. If you need to take time off, let us know if we're close to deadline; otherwise, just let us know when you'll be back. If it's an emergency, leave a note & go; don't fret about it. Keep in touch while you're gone." Funniest darned thing - in 20 years, I lost 3 to quitting; 2 because their wives got really good jobs too far away for them to commute & 1 who, at the tender age of 63, was inspired by my methods to open his own shop. He was sad about us having to compete until I told him, "Hank, there will be times when you will need work & we'll need another subcontractor. And there may be times when we will need work & you will need some help. We don't 'have' to eat each other to stay alive." He left a happy man, as did the other 2. In those same 20 years, I had to fire 3; 2 for closet alcoholism & 1 for trying to sabotage the shop. The saboteur cost us an $85,000 commission in November when work would get scarce. That commission would have kept us all fed & warm thru the winter. Not only did I fire him, but as the President of a 6-state regional professional group, I notified every similar shop within 150 mile radius of us of his antics, & last anyone heard, he was stocking shelves at WalMart. He was let go in front of the entire crew, & they all knew why, because I have never been one to mince words. When I went to Chamber of Commerce meetings, literally everyone else would be wailing about how they couldn't get good help. I obviously never had that problem & the reason was simple - I treated my people as intelligent capable ADULT human beings & did everything I could to keep them happy because... (Duh-uh-uh..) HAPPY PEOPLE WORK HARDER & PRODUCE BETTER RESULTS. Funy thing is, it isn't hard to do because most people's wants are simple. This didn't need a study by a major university to figure out. It is basic how-to-do.

Hell is a strong word, but that's what Us chooses to describe Heidi Klum's marriage to Seal, which is coming to an end after nearly seven years of bliss together.

While reconciliation is still on the table, and the couple is still wearing their wedding rings, the tabloid tells a dramatically different story regarding the duo.

One focused on Seal's temper, apparently.

While jealousy over Heidi's career was reportedly a source of friction between the two, as well as the time they spent apart, "vicious fights" may be a stretch.

Merle Haggard returns to Calif. home to recuperate (AP)

NASHVILLE, Tenn. ? Country music legend Merle Haggard has returned home to northern California to recuperate from multiple illnesses.

Haggard had been hospitalized in Macon, Ga., for about a week with double pneumonia. While there, doctors discovered three stomach ulcers and eight polyps in his colon.

At one point, the 74-year-old checked himself out of the hospital to return to Redding, Calif., by tour bus. But he returned a few hours later after deciding the trip would be too taxing.

The "Okie from Muskogee" singer was diagnosed with lung cancer a few years ago, but recovered after part of his lung was removed.

It's unclear how Haggard's recent health issues will affect his touring schedule. His website shows him playing next on Feb. 28 in Tucson, Ariz.

Spinoff for "The Office" in the works at NBC (Reuters)

LOS ANGELES (Reuters) ? Quirky workplace TV comedy "The Office" may be headed for greener pastures.

A source close to the NBC show said on Thursday that the network was working on a possible spinoff involving actor Rainn Wilson's character Dwight Schrute going back to his family's farm.

No deals have been signed yet and NBC declined to comment.

But industry website Deadline Hollywood said the proposed show would see Dwight, who was dismayed at being passed over for the job of Dunder Mifflin office manager last year, living at his family's beet farm and bed and breakfast. The show could include multiple generations of his family struggling to survive and stay together.

Deadline said that if all goes well, the new show could be launched in early 2013.

"The Office", based on the British mockumentary of the same name created by comic Ricky Gervais, has been a critical and ratings success for NBC for eight years, winning Emmys, acting and writing awards.

But ratings have slipped by about one million regular viewers since the departure last year of lead actor Steve Carell, who portrayed manager Michael Scott.

Demi Moore's Hospitalization: More Details Emerge

TMZ reports that whip-its were behind actress' health scare, while People says friends have been worried about Moore for a while.

While the many reported reasons for Demi Moore's recent hospitalization have varied from exhaustion to substance abuse, TMZ is now saying nitrous oxide was behind the health scare.

According to the website, friends who were on the scene the night of the hospitalization told EMTs the actress was inhaling nitrous oxide from aerosol cans — also known as whip-its — before 911 was called. TMZ's sources said the actress was exhibiting seizure-like symptoms after inhaling the nitrous.

Demi's hospitalization caps a rough patch for the actress.

People magazine also talked to sources close to Moore, who said they've been worried about their friend for quite some time. "Really, it was over the last year her friends saw a change," one source said about her split from husband Ashton Kutcher. "She wasn't sleeping as well, didn't seem to be eating and looked really gaunt."

Another source told the mag that Moore's recent prescription-drug abuse was a factor in the end of her marriage. "It was a sticking point for Ashton," the source said. "He wanted her to take care of herself and get a hold of things, and she wouldn't."

Moore endured a very public and emotional split from Kutcher back in November, publicly announcing the breakup with "sadness and a heavy heart" after months of not-so-quiet rumblings that her husband had been unfaithful. While it was announced earlier this month that Moore would star as feminist icon Gloria Steinem in the upcoming film "Lovelace," her rep said the setback in her health would prevent her from working on the project.

Muslims urge resignation of NY police chief over video (Reuters)

NEW YORK (Reuters) ? U.S. Muslim civil rights groups demanded the resignation of New York Police Commissioner Ray Kelly on Wednesday amid a controversy over the repeated screening of an offensive video.

Kelly said he regretted cooperating with the makers of "The Third Jihad: Radical Islam's Vision for America," which shows footage of suicide attacks and says "the true agenda of much of Muslim leadership here in America" is to "infiltrate and dominate America."

Kelly came under fire following reports that the video had been screened many more times than previously acknowledged. When the video first came to light a year ago, police said it had been screened only a few times.

In fact, it was shown to more than 1,400 officers over a period of months, the New York Times reported on Tuesday based on documents obtained in a Freedom of Information Act request.

The Council on American-Islamic Relations (CAIR), a prominent Muslim civil liberties organization, said Kelly had disqualified himself to head the country's largest and most prominent police force.

"As leaders of the nation's largest police department, Commissioner Kelly and Deputy Commissioner (Paul) Browne's actions set a tone for relations with law enforcement that impact American Muslims nationwide," CAIR National Executive Director Nihad Awad said in a statement. "It's time for change."

CAIR and other civil liberties groups set a news conference for Thursday at New York City Hall.

The controversy comes as Kelly, who is closely aligned with Mayor Michael Bloomberg, was restoring the police department's strained relations with the Muslim minority.

"Somebody exercised some terrible judgment," Bloomberg told reporters on Tuesday, referring to the film. "As soon as they found out about it, they stopped it."

STRAINED RELATIONS

"The Third Jihad" ran on continuous loop on a TV in a Brooklyn police location that officers used to fill in paperwork during down time, Kelly's spokesman and deputy, Paul Browne, said. He said the film was not used in training sessions and was never shown at the Police Academy.

In addition, police now admit a spokesman helped arrange an interview the filmmakers did with Kelly that appeared in the film. Previously, police had said Kelly was not involved in the making of the film and that the interview was taken from an archive.

Browne said on Wednesday the commissioner finds the finished product "objectionable" and regrets taking part.

The filmmakers did not immediately respond to a request for comment.

Kelly has navigated difficult times with Muslims, having come into office shortly after the September 11 attacks of 2001, which led to an unprecedented security crackdown that drew the ire of civil libertarians.

Kelly and Bloomberg have since won praise for prosecuting hate crimes against Muslims and defending the right to build a mosque near the site of the September 11 attacks. But there have been periods of tension, more recently over the department's role in secret operations at New York area mosques.

Wikileaks announces Julian Assange TV show, world governments fire up their DVRs

Ready or not, Julian Assange is heading toward a TV set near you. Wikileaks announced this week that its controversy-embroiled founder will be getting his own TV show, in which he'll be interviewing "key political players, thinkers and revolutionaries." Information on the series is light at present -- the largely unknown Quick Roll Productions will apparently play a role in its creation. The show is set to begin airing in the middle of March and will run as ten 30 minute weekly episodes. Assange for one, clearly has grand ambitions for the series, stating that it "will explore the possibilities for our future in conversations with those who are shaping it. When XDA Developers member verygreen came across the pleas of one user obsessed with attaching an external USB keyboard to an eReader, he did what any decent hacker would and created a workable solution. Using a loaned Nook Simple Touch, this self-styled Make-A-Wish Hack was able to patch Barnes & Noble's existing kernel, which already supported USB host mode, and send commands over ADB to enable the connection. It's not a foolproof workaround, though, as only low-power devices will function without additional juice and even so, at a great cost to the greyscale device's battery life. Sure, this may not excite you much, but it's certainly made for one very satisfied forum dweller out there in cyberland. After all, isn't that what haxxors are for?
